A new way to treat colon cancer?
Scientists at University of Utah's Huntsman Cancer Institute have discovered a new target for possible future colon cancer therapys a molecule that is implicated in 85 percent of colon cancer cases.

These findings were published online Oct. 6, 2006, in the Journal of Biological Chemistry.

By knocking out that is, genetically disabling a molecule called C-Terminal Binding Protein (CTBP) scientists were able to rescue zebrafish from the effects of a mutation in the adenomatous polyposis coli (APC) gene.

In humans, mutations in this gene long have been known to initiate a series of events that cause colon polyps, which eventually become malignant. APC mutations play a role in 85 percent of colon cancers. The new findings mean CTBP also is involved in that proportion of colon cancers.

In zebrafish, APC mutations keep the intestine from developing properly. "In essence, knocking out CTPB promotes normal development of the intestine in zebrafish carrying an APC mutation," says David A. Jones, a University of Utah associate professor of oncological sciences and leader of the study.

In normal cells of both humans and zebrafish, the APC gene controls the amount of CTBP present by marking it for destruction. In tumor cells with mutated APC, CTPB is not destroyed; instead it accumulates in the cell.........

Scientists in the United Kingdom have developed a new wound dressing that could bring the benefits of maggot therapy to patients without putting live Greenbottle fly (blowfly) larvae into non-healing wounds. The joint research project of Stephen Britland from Bradford University and David Pritchard of Nottingham University included colleagues from the Bradford-based biotechnology company AGT Sciences Ltd. It describes development and preliminary testing in laboratory cell cultures of the new hydrogel dressing in a report scheduled for publication in the Oct. 6 issue of the ACS bimonthly journal Biotechnology Progress.

The researchers note resurgence in medical use of larval biotherapy -- intentionally introducing blowfly maggots into non-healing wounds to clean away dead tissue. Medical use of the technique led to observations suggesting that maggots' excretions and secretions (ESs) also may encourage regeneration of tissue and wound healing. Realizing that the ESs would have to be delivered in a controlled fashion, Britland's group developed the hydrogel dressing, which slowly releases maggot ESs.

"The present prototype hydrogel wound dressing could potentially be deployed as a device to deliver insect-derived active products to skin wounds in vivo to encourage tissue regeneration".........

Many women carry a private sorrow. They suffer from uneven breasts; with each breast differing is size from the other. This condition is also known as breast asymmetry and is a relatively common condition. The embarrassment about the uneven breasts could affect their daily lives, sexuality and confidence. Researchers say that for those women with significant asymmetry, breast surgery can considerably elevate quality of life and self-esteem. These discussions were part of as study that was presented recently at the American Society of Plastic Surgeons (ASPS) Plastic Surgery 2006 conference in San Francisco.

"All women have some degree of breast asymmetry, but for those with a noticeable difference, the embarrassment often keeps them from seeking help," said Walter Erhardt, MD, ASPS Member Surgeon and Public Education Committee chair. "Even if breasts differ by less than a half-cup size it can be very noticeable. The condition is talked about so infrequently that a number of are unaware there are surgeries that can correct the problem".

Breast asymmetry occurs when a woman's breasts differ in size or shape. For example, a woman's left breast may be a B-cup and the right, a D-cup. Conversely, a woman may have one breast that significantly droops, and the other does not, making the woman's breasts appear unbalanced.........

Cola Might Increase Osteoporosis Risk
According to the National Osteoporosis Foundation, approximately 55 percent of Americans, mostly women, are at risk of developing osteoporosis, a disease of porous and brittle bones that causes higher susceptibility to bone fractures. Now, Katherine Tucker, PhD, director of the Epidemiology and Dietary Assessment Program at the Jean Mayer USDA Human Nutrition Research Center on Aging at Tufts University, and colleagues have reported findings in the American Journal of Clinical Nutrition that cola, a popular beverage for many Americans, may contribute to lower bone mineral density in older women, a condition which increases risk for osteoporosis.

Tucker, also a professor at the Friedman School of Nutrition Science and Policy at Tufts, and colleagues analyzed dietary questionnaires and bone mineral density measurements at the spine and three different hip sites of more than 2,500 people in the Framingham Osteoporosis Study whose average age was just below 60. In women, cola consumption was associated with lower bone mineral density at all three hip sites, regardless of factors such as age, menopausal status, total calcium and vitamin D intake, or use of cigarettes or alcohol.

However, cola consumption was not associated with lower bone mineral density for men at the hip sites, or the spine for either men or women. The results were similar for diet cola and, although weaker, for decaffeinated cola as well.........

Tumor Suppressor Promoteing Cancer Cell Growth?
Scientists have shown that the tumor suppressor gene H-REV107-1 may actually stimulate tumor progression in some non-small cell lung carcinomas. The related report by Nazarenko et al., "H-REV107-1 stimulates growth in non-small cell lung carcinomas via the activation of mitogenic signaling," appears in the recent issue of The American Journal of Pathology.

Tumor suppressor genes function by regulating normal cell growth and proliferation. When a tumor suppressor gene is turned off, by mutation, deletion, or blocked expression, cell growth can proceed without safeguards, contributing to cancer cell proliferation. However, this appears not to be the case in some non-small cell lung carcinomas (NSCLC), in which a tumor suppressor (H-REV107-1) actually promotes cancer cell growth.

Nazarenko et al. found H-REV107-1 expression in a portion of human NSCLC samples examined. When they further characterized this expression in relation to normal lung tissue, H-REV107-1 was found in nonproliferating and proliferating cells in normal lung tissue, localized mainly to the nucleus. In cultured NSCLC cells, however, H-REV107-1 was found in either the cytoplasm or both the cytoplasm and nucleus.

The group then examined whether cellular localization of H-REV107-1 in NSCLC tumor samples is linked with tumor behavior. Strikingly, cytoplasmic localization correlated with decreased patient survival (24 months versus 41 months for nuclear localization). These data suggested that cytoplasmic H-REV107-1 stimulates cell growth. This was then confirmed by suppression of H-REV107-1 RNA, which inhibited cell proliferation, and overexpression of H-REV107-1 protein, which stimulated cell growth pathways and increased proliferation.........

Research published recently in Journal of the CardioMetabolic Syndrome (JCMS) presents data supporting that adult diseases, such as hypertension, type 2 diabetes, and sleep apnea are now recognizable in childhood. The underlying link between them is a disorder of insulin resistance, which is worsened by childhood obesity. The annual National Health and Nutrition Examination Survey by the Centers for Disease Control and Prevention observed that about one-third of U.S. children today, about 25 million, are overweight or at risk of becoming overweight.

One study in this special issue reports data on the effect of age and sex on cardiovascular risk in overweight children, aged 11 years and older. Results showed that hypertension and dyslipidemia in overweight children is high, with overweight males 11 years and older having a higher prevalence of these risk factors than females and younger males. This may explain the earlier appearance of cardiovascular disease in overweight adult males.

Scientists from Wisconsin, lead by Dr. David K. Murdock examined the effect of elevated body mass index in 247 healthy school children of which 28 percent of 2nd graders and 33 percent of 11th graders were overweight. Data from the study revealed that biomarkers of increased risk of adverse cardiovascular outcomes were already present in the overweight children.........

Opening a new front in the war against flu, scientists at the University of Wisconsin-Madison have reported the discovery of a novel compound that confers broad protection against influenza viruses, including deadly avian influenza.

The new work, reported online this week in the Journal of Virology, describes the discovery of a peptide -- a small protein molecule -- that effectively blocks the influenza virus from attaching to and entering the cells of its host, thwarting its ability to replicate and infect more cells.

The new finding is important because it could make available a class of new antiviral drugs to prevent and treat influenza at a time when fear of a global pandemic is heightened and available antiviral drugs are losing their potency.

"This gives us another tool," says Stacey Schultz-Cherry, a UW-Madison professor of medical microbiology and immunology and the senior author of the new report. "We're quickly losing our antivirals".

The new drug, which was tested on cells in culture and in mice, conferred complete protection against infection and was highly effective in treating animals in the early stages of infection. Untreated infected animals typically died within a week. All of the infected animals treated with small doses of the drug at the onset of symptoms survived.........

Researchers found that curcumin -- a chemical found in curry and turmeric -- may help the immune system clear the brain of amyloid beta, which form the plaques found in Alzheimer's disease.

Published in the Oct. 9 issue of the Journal of Alzheimer's Disease, the early laboratory findings may lead to a new approach in treating Alzheimer's disease by enhancing the natural function of the immune system using curcumin, known for its anti-inflammatory and anti-oxidant properties.

Using blood samples from six Alzheimer's disease patients and three healthy control patients, the researchers isolated cells called macrophages, which are the immune system's PacMen that travel through the brain and body, gobbling up waste products, including amyloid beta.

The team treated the macrophages with a drug derived from curcumin for 24 hours in a cell culture and then introduced amyloid beta. Treated macrophages from three out of six Alzheimer's disease patients showed improved uptake or ingestion of the waste product compared to the patients' macrophages not treated with curcumin. Macrophages from the healthy controls, which were already effectively clearing amyloid beta, showed no change when curcumin was added.

"Curcumin improved ingestion of amyloid beta by immune cells in 50 percent of patients with Alzheimer's disease. These initial findings demonstrate that curcumin may help boost the immune system of specific Alzheimer's disease patients," said Dr. Milan Fiala, study author and a researcher with the David Geffen School of Medicine at UCLA and the VA Greater Los Angeles Health Care System. "We are hopeful that these positive results in a test tube may translate to clinical use, but more studies need to be done before curcumin can be recommended".........

Children with Williams syndrome, a rare genetic disorder, just love music and will spend hours listening to or making music. Despite averaging an IQ score of 60, a number of possess a great memory for songs, an uncanny sense of rhythm, and the kind of auditory acuity, than can discern differences between different vacuum cleaner brands.

A study by a multi-institutional collaboration of scientists, published in a forthcoming issue of NeuroImage, identified structural abnormalities in a certain brain area of people afflicted with Williams syndrome. This might explain their heightened interest in music and, in some cases, savant-like musical skill.

Professor Ursula Bellugi, director of the Laboratory for Cognitive Neuroscience at the Salk Institute for Biological Studies the central hub of this unique scientific alliance explains, "Understanding the connections between missing genes, the resulting changes in brain structure and function, and ultimately behavior may help us to reveal how the brain works".

The current study is just the latest chapter in a story that's been unfolding for quite some time gaining increasing momentum in recent years. It all started when Bellugi reached out across disciplines and assembled a team of experts under the umbrella of a Program Project from the National Institutes of Child Health and Human Development to help her trace the influence of individual genes on the development and functioning of the brain.........

We all know about the flu-vaccine shortage that occurred during the last flu season. Scientifics are formulating strategies to combat flu vaccine shortage in case of shortage during the peak flu season. Public health officials are now making mathematical model at The University of Texas at Austin to determine the best way to distribute the vaccine.

The scientists used contact network epidemiology to model various vaccine distribution strategies, including the United States Centers for Disease Control strategy of targeting high-risk groups, like infants, the elderly and people with health complications. They also tested the idea of targeting school children, who are critical vectors in moving diseases through communities.

They found that the best vaccine distribution strategy depends on the contagiousness of the flu strain.

"If we only have a limited flu vaccine supply, the best distribution strategy depends on the contagiousness of the strain," says Dr. Lauren Ancel Meyers, assistant professor of integrative biology. "We can more effectively control mildly contagious strains by vaccinating school children, while we can more effectively control moderately and highly contagious strains by vaccinating high-risk groups".

If there is no information available about the contagiousness of a flu strain or if the vaccines are only available after the outbreak is underway, the study recommends prioritizing vaccines for those people in high-risk groups who can experience the greatest complications due to the disease.........
