A new study by scientists at the MUHC has revealed that a diet low in folate may increase the risk of developing colorectal cancer. Published in the scientific journal Cancer Research today, the study not only illustrates a way to prevent the disease but also provides further insight into the mechanisms of the disease, which could lead to novel therapies. Using animal models, the MUHC study is the first to demonstrate directly that diets low in folate cause colorectal cancer, and follows on the heels of earlier research by the same team that revealed how high folate diets can protect against heart disease.

"This research, which is consistent with previous epidemiological studies in humans, demonstrates a clear link between low dietary folate and the initiation of colorectal cancer in animal models," says Dr. Rima Rozen, Scientific Director of the Montreal Children's Hospital, Deputy Scientific Director of the MUHC, and lead investigator in the study. "None of the mice fed a control diet developed tumours whereas 1 in 4 mice on the folate-deficient diet developed at least one tumour".

Possible mechanisms of the disease were also investigated using molecular biological techniques. "The study shows that a low level of dietary folate may cause an increase in DNA damage, which plays a role in the development of tumours," noted Dr. Rozen. The study also reveals that folate deficiency causes genes which usually respond to DNA damage to behave abnormally, also contributing to the development of tumours. The results suggest that a diet containing sufficient folate may reduce the risk of developing colorectal cancer.........

Fundamental theories regarding consciousness, emotion and quality of life in sufferers of paralysis from Amyotrophic Lateral Sclerosis (ALS, also known as 'Lou Gerhig's disease') are being challenged based on new research on brain-computer interaction. ALS is a progressive disease that destroys neurons affecting movement. The study appears in the latest issue of Psychophysiology. The article reviews the usefulness of currently available brain-computer -interfaces (BCI), which use brain activity to communicate through external devices, such as computers.

The research focuses on a condition called the completely locked-in state (CLIS, a total lack of muscle control). In a CLIS situation, intentional thoughts and imagery can rarely be acted upon physically and, therefore, are rarely followed by a stimulus. The research suggests that as the disease progresses and the probability for an external event to function as a link between response and consequence becomes progressively smaller, it may eventually vanish altogether.

Researchers have found that by implementing a BCI before the CLIS state occurs, a patient can be taught to communicate through an electronic device with great regularity. The continued interaction between thought, response and consequence is believed to slow the destruction of the nervous system.........

Alzheimer's Drug Shows Promise
The only drugs currently available for Alzheimer's patients are those that alleviate symptoms, but a team of researchers led by Paul Aisen, MD, director of the Memory Disorders program at Georgetown University Medical Center, is testing a new class of drugs that actually target the molecule believed to cause the disease. Aisen and colleagues report that a compound called tramiprosate reduced levels of a marker for the progression of Alzheimer's disease in a Phase II clinical trial in the November 1 electronic version of Neurology.

"Everyone wants to figure out how to create an Alzheimer's therapy that attacks the amyloid peptide, which is considered to be the molecular cause of the disease," said Aisen. "This is the most advanced anti-amyloid therapy that exists-it has the potential for slowing down progression of the disease".

Aisen and his team are currently in the midst of Phase III clinical trials on tramiprosate (manufactured by Neurochem, for which Aisen is a scientific advisor, as ALZHEMED-) and hope to have results by early next summer. MRI Study To Prevent Brain Damage
A stroke victim arrives in the emergency room and, within minutes, the doctor must make a decision: Should drugs be administered to open up the blocked blood vessel and prevent further brain damage? Or is this patient at high risk for suffering a brain hemorrhage if the blocked vessel is opened?

Greg Albers, MD, director of the Stanford Stroke Center, and his team report in the recent issue of Annals of Neurology that new magnetic resonance imaging techniques can discriminate between stroke patients who are likely to benefit from a stroke medicine - even when administered beyond the currently approved three-hour time window - and those for whom treatment is unlikely to be beneficial and may cause harm.

For years, Albers, professor of neurology and neurological sciences at the Stanford University School of Medicine, has been using new MRI techniques to visualize the damage from stroke while it is actually happening. His goal is to differentiate brain tissue that is potentially salvageable from tissue that is already irreversibly injured by a stroke. As his group accumulated MRI scans of stroke patients, they noticed patterns that seemed to identify which patients were most likely to benefit from opening up blocked blood vessels.

"One of the criticisms was that these detailed brain images looked beautiful and interesting, but there was no proof that they should be used to influence treatment or that they would result in improved outcomes," said Albers. "How do you know that these MRI patterns can predict whether the therapy is likely to be beneficial?".........

A research team found that when baby rhesus monkeys endured high rates of maternal rejection and mild abuse in their first month of life, their brains often produced less serotonin, a chemical that transmits impulses in the brain. Low levels of serotonin are associated with anxiety and depression and impulsive aggression in both humans and monkeys.

Abused females who became abusive mothers in adulthood had lower serotonin in their brains than abused females who did not become abusive parents, the research showed.

Because the biological make up of humans and monkeys is quite similar, the findings from the monkey research could be valuable in understanding human child abuse, said Dario Maestripieri, Associate Professor in Comparative Human Development at the University of Chicago.

"This research could have important implications for humans because we do not fully understand why some abused children become abusive parents and others don't," Maestripieri said. The research suggests that treatments with drugs that increase brain serotonin early in an abused child's life could reduce the likelihood that the child will grow up to become abusive, Maestripieri said.

Maestripieri is lead author of a paper reporting the research, "Early Maternal Rejection Affects the Development of Monoaminergic Systems and Adult Abusive Parenting in Rhesus Macaques" published in the current issue of Behavioral Neuroscience.........

Poor Readers Have Higher Risk Of Suicide
Teenagers with reading problems are at significantly higher risk for suicide and for dropping out of school than typical readers, according to a study by Wake Forest University Baptist Medical Center researchers.

"In our study, poor readers were three times more likely than typical readers to consider or attempt suicide and six times more likely to drop out of school," said lead author Stephanie Sergent Daniel, Ph.D. "Educators and parents should be aware of the risk of suicidal thoughts and behavior among adolescents with reading problems".

The results, reported today in the recent issue of the Journal of Learning Disabilities, are from a study of 188 students recruited from six public high schools at age 15. They were followed for a mean of 3.3 years.

Researchers initially screened 1,074 students and identified a sub-group willing to participate in the long-term study. From this group, they recruited a group of poor readers and a group of typical readers that were matched for gender and race.

Standard educational tests were used to measure single-word reading ability, one of several skills involved in reading. Students scoring in the lowest 18 percent were considered poor readers - a cutoff commonly used to diagnose dyslexia. In addition, each student and his primary caretaker were interviewed by master's level trained research clinicians to assess psychiatric disorders and suicidal behaviors. The median length between interviews for students and parents was twelve months.........

Antidepressants Linked To Lower Child Suicide Rates
Researchers report an inverse relationship between antidepressant prescriptions and the rates of suicide in children and adolescents -- a finding that contradicts the Food and Drug Administration's "black box" warning for selective serotonin reuptake inhibitor medications, also known as SSRI drugs.

The University of Illinois at Chicago epidemiologic study appears in the recent issue of the American Journal of Psychiatry.

The researchers examined suicide rates of children ages 5-14 in each county of the United States from 1996 to 1998 and county-level data on SSRI prescriptions. The results were adjusted for sex, race, income, access to quality mental health care and variations in county-to-county suicide rates.

"We found that counties with the highest prescription rates for SSRI drugs had the lowest suicide rates in children and adolescents," said the lead author Robert Gibbons, director of the Center for Health Statistics and professor of biostatistics and psychiatry at UIC. "This is just the opposite of what you would predict if SSRI's were producing suicide".

There were 933 suicides among children ages 5-14 from 1996 to 1998, or an overall annual rate of 0.8 per 100,000. The researchers found that in counties with low antidepressant prescription rates, the suicide rate was as high as 1.7 per 100,000. In counties with high antidepressant prescription rates, the suicide rate was as low as 0.7 per 100,000.........

A study released recently by the University of Minnesota's "Project EAT" (Eating Among Teens) shows startling results of 2,500 female teenagers studied over a five-year period. The study found that high school-aged females' use of diet pills nearly doubled from 7.5 to 14.2 percent. By the ages of 19 and 20, 20 percent of females surveyed used diet pills.

"These numbers are startling, and they tell us we need to do a better job of helping our daughters feel better about themselves and avoid unhealthy weight control behaviors," U of M professor and study researcher Dianne Neumark-Sztainer said.

Other results from the study include:

62.7 percent of teenage females use "unhealthy weight control behaviors"

21.9 percent of teenage females use "very unhealthy weight control behaviors"

Very unhealthy weight control behaviors include the use of diet pills, laxatives, vomiting or skipping meals. Of the 2,500 teenage males studied, their rates were half of the females'.

"We have found that teenage females who diet and use unhealthy weight control behaviors are at three times the risk of being overweight," said Neumark-Sztainer. "Teens who feel good about their bodies eat better and have less risk of being overweight. Parents can play a key role in helping their children to build a positive body image and engage in healthy eating and physical activity behaviors".........

Staph Vaccine Shows Promise Staphylococcus aureus
By combining four proteins of Staphylococcus aureus that individually generated the strongest immune response in mice, scientists have created a vaccine that significantly protects the animals from diverse strains of the bacterium that cause disease in humans. A report describing the University of Chicago study, funded by the National Institute of Allergy and Infectious Diseases (NIAID) of the National Institutes of Health, appears online this week in the Proceedings of the National Academy of Sciences.

"This finding represents a promising step toward identifying potential components to combine into a vaccine designed for people at high risk of invasive S. aureus infection," notes Anthony S. Fauci, M.D., NIAID director.

S. aureus, the most common agent of hospital-acquired infection, is the leading cause of bloodstream, lower respiratory tract and skin infections. These infections can result in a variety of illnesses, including endocarditis (inflammation of the heart), toxic-shock syndrome and food poisoning.

Research in S. aureus has taken on new urgency: In the past few decades, the bacterium has developed resistance to traditional antibiotics, thus allowing infections to spread throughout the body of the infected individual despite treatment. More recently, healthy people with no apparent risk factors have been infected by novel and extremely virulent strains of S. aureus acquired from community rather than hospital sources.........

Speaking In Tongues

Speaking In Tongues
Glossolalia, otherwise referred to as "speaking in tongues," has been around for thousands of years, and references to it can be found in the Old and New Testament. Speaking in tongues is an unusual mental state associated with specific religious traditions. The individual appears to be speaking in an incomprehensible language, yet perceives it to have great personal meaning. Now, in a first of its kind study, scientists are shining the light on this mysterious practice -- attempting to explain what actually happens physiologically to the brain of someone while speaking in tongues.

Researchers at the University of Pennsylvania School of Medicine have discovered decreased activity in the frontal lobes, an area of the brain associated with being in control of one's self. This pioneering study, involving functional imaging of the brain while subjects were speaking in tongues, is in the recent issue of Psychiatry Research: Neuroimaging, the official publication of the International Society for Neuroimaging in Psychiatry.

Radiology scientists observed increased or decreased brain activity - by measuring regional cerebral blood flow with SPECT (Single Photon Emission Computed Tomography) imaging - while the subjects were speaking in tongues. They then compared the imaging to what happened to the brain while the subjects sang gospel music.........
